Do I need to add flux to unfluxed desolder braid?

Yes, without flux, solder will not flow and wick up the braid so you must add flux. The appeal of unfluxed braid is you can add your own flux.

Do I need to remove any conformal coating before using the desolder braid?

Yes, the high temperatures used to melt the solder would burn the conformal coating and release toxic fume, therefore it is strongly recommended that any conformal coating be removed prior to removing solder from the joints.
